there's no big hills to speak of, though the end is a bit up-and-down and that can sap your energy a bit...
Are there many hills? I only ride around London where it is flat. How much harder is it than say the London to Brighton ride?
When we did it I think we left at 8.30 and got there about 7 or so the next morning, including a hour stop in the middle...
I must remember to charge my lights up! How long does it normally take riding on one gear?
You'll be far from alone doing it fixed. My friend Tim and I will be there, he fixed, me just single-speed... did it two years ago and it was a catalogue of pain but brilliant nonetheless. Get yourself some serious lights and pray for a rain-free night...
